ABSTRACT
2′,4′-Dihydroxychalcone (2′,4′-DHC) was identified from a heat shock protein 90 (Hsp90)-targeting library as compound with Hsp90 inhibitory and antifungal effects. In the presence of 2′,4′-DHC (8 μg/mL), radial growth Aspergillus fumigatus inhibited 20% compared to control, green pigmentation completely blocked. The expression conidiation-associated genes abaA, brlA, wetA significantly decreased (approximately 3- 5-fold) by treatment 2′,4′- DHC. calcineurin signaling components, cnaA crzA, also reduced. effects on metabolic activity mycelial were enhanced combination itraconazole caspofungin. Docking studies indicated that bind ATPase domain Hsp90. These results suggest act an Hsp90-calcinurin pathway inhibitor.
