Roles of the Bcl-2/Bax Ratio, Caspase-8 and 9 in Resistance of Breast Cancer Cells to Paclitaxel

ABSTRACT
The goal of this study was to establish paclitaxel resistant MCF-7 cells, as in vitro model, identify the molecular mechanisms leading acquired chemoresistance breast cancer cells. Resistant cells were developed by stepwise increasing exposure paclitaxel. Gene expression levels Bax and Bcl-2 along with protein caspase-8 caspase-9 evaluated two cell lines (MCF-7/Pac64 MCF-7/Pac5 nM). Morphological modifications resistance examined light microscopy fluorescence activated sorting (FACS). As an important indicator chemotheraputic agents, Bcl-2/Bax ratio showed a significant increase both MCF-7/Pac5nM MCF-7/Pac 64nM (p<0.001), while decreased (p<0.001) increased (p<0.001). FACS analysis demonstrated that MCF-7/Pac64 smaller than no difference their granularity. Our results support idea induces apoptosis mitochondrial-dependent manner. Identifying patients higher caspase 9 level then inhibiting activity these proteins may improve efficacy agents.
