ABSTRACT
Biological and pharmacological activities of dryocrassin ABBA, a phloroglucinol derivative extracted from Dryopteris crassirhizoma, have attracted attention. In this study, the apoptotic effect ABBA on human hepatocellular carcinoma HepG2 cells was investigated.We tested effects in vitro by MTT, flow cytometry, real-time PCR, Western blotting. KM male mice were used to detect H22 vivo.Dryocrassin inhibited growth concentration-dependent manner. After treatment with 25, 50, 75 μg/mL cell viability 68%, 60% 49%, respectively. Dryocrassin able induce apoptosis, measured propidium iodide (PI)/annexin V-FITC double staining. The results PCR ting showed that up-regulated p53 Bax expression Bcl-2 which led an activation caspase-3 caspase-7 cytosol, then induction apoptosis. vivo experiments also significantly suppressed tumor growth, without major side effects.Overall, these findings provide evidence may apoptosis through caspase-mediated mitochondrial pathway.
