ABSTRACT
To explore the mechanism of ABO discrepancy in a patient by genotyping and reasonable blood transfusion strategy.Routine serological test was carried out to analyze group. The presence group determinants on red cells were determined adsorption-elution test. Exons 1-7 adjacent introns gene amplified PCR sequenced. The showed forward reverse typing discrepancy. defined as B, however, indicated that AB subtype. Absorption-elution confirmed weak A antigens patient's cells. sequencing T>C exchange at position exon 7 which resulted isoleucine threonine substitution codon 256. genotype ABO*Ael05/B101.The 767 α-1,3-N-acetyl galactose is molecular leading decrease expression antigen Ael05
