ABSTRACT
Research in the field of human immunology is restricted by lack a system that reconstitutes in-situ activation dynamics quiescent antigen-specific T-cells interacting with dendritic cells. Here we report tissue-like recapitulates engineered primary immune cell. Our approach facilitates real-time single-cell manipulations, tracking interactions and functional responses complemented population-based measurements cytokines, status proliferation. As proof concept, recapitulate immunological phenomenon such as CD4 T-cells' help to CD8 through enhanced maturation DCs effect PD-1 checkpoint blockades. In addition, characterise unique T-cell/DC function antigen affinity.
