ABSTRACT
Wnt signaling plays a crucial role in the early embryonic patterning and development, to regulate convergent extension during gastrulation establishment of dorsal axis. Further, is regulator craniofacial morphogenesis. The adapter proteins Dact1 Dact2 modulate pathway through binding Disheveled. However, distinct relative functions embryogenesis remain unclear. We found that dact1 dact2 genes have dynamic spatiotemporal expression domains are reciprocal one another wnt11f2 , suggest zebrafish embryogenesis. both contribute axis extension, with compound mutants exhibiting similar defect phenotype mutant. Utilizing single-cell RNAseq gpc4 mutant disrupts noncanonical signaling, we identified dact1/2 specific roles development. Comparative whole transcriptome analysis between wildtype, revealed novel for regulating mRNA classical calpain capn8 . Over-expression phenocopies dysmorphology. These results identify previously unappreciated calcium-dependent proteolysis Taken together, our findings highlight overlapping providing new insights into multifaceted regulation signaling.
