This paper describes a new cell isolation and HLA typing technique, which permits separation class I or II to be performed in 70 min. Magnetic monodisperse microspheres (Dynabeads TM) were coated with monoclonal antibodies (MAbs) specific for the CD8 T antigen monomorphic epitopes. They could then used obtain positive cells directly from ACD blood approximately 15 min by use of magnetic separation. The (attached microspheres) subsequently microcytotoxic (total incubation time 55 min) using...
The terminal complement complex (TCC), consisting of C5b, C6, C7, C8, and C9, contains neoantigens that are absent from the individual native components. Neoantigens present both in membrane‐bound (MAC) fluid‐phase (SC5b‐9) complex. study describes production monoclonal antibodies against forms TCC. A convenient screening detection system, based mainly on enzyme‐linked immunosorbent assays, crossed immunoelectrophoresis with autoradiography, affinity chromatography subsequent sodium dodecyl...
Activation products of the complement cascade contain neoepitopes that are not present in individual native components. Monoclonal antibodies detecting have been used for direct quantification activation at different steps cascade. These methods suggested to be more sensitive and reliable than conventional tests, which hampered by precipitation or fractionation procedures. The study describes production screening characterization a monoclonal antibody (MoAb) bH6, MoAb bH6 exhibited...
Abstract The principle of activated swelling has made available polymer particles in the range 1–100 μm, highly uniform size, and with great variations morphology chemical composition. A short description method is given. On basis monosized porous macroreticular particles, magnetic (MMP) are prepared which size as well content oxides. These have found applications a number selective cell separation procedures. Special types MMP been developed for isolation organelles viruses. use contrast...
Routine one-step centrifugation procedures (Lymphoprep = LP, Percoll) commonly used for separation of blood cells split the into two major fractions. After mononuclear (MNC monocytes and lymphocytes) are located on top fluid, whereas erythrocytes granulocytes have sedimented to bottom. We now show that a relatively pure lymphocyte suspension can be obtained by citrated using NycoPrep (NP iohexol), nonionic X-ray contrast agent. With this gradient medium also pass bottom, leaving lymphocytes...
Summary: IgG subclass concentrations were determined in sera from 20 epileptic patients before carbamazepine therapy and after 6 weeks of treatment. Subclass‐specific monoclonal antibodies used an ELISA technique. Carbamazepine reduced the IgG2 concentration 13 patients, mean value fell 3.21 to 2.47 g/L during treatment (p < 0.01). This decrease was maintained 4 12 months therapy. IgGl, IgG3, IgG4 did not change. deficiency seen. The reduction related serum within therapeutic range, type...
This study investigated the phenotype of freshly isolated human tumour infiltrating lymphocytes (TIL) from 14 patients with colorectal tumours, and compared them derived lamina propria unaffected mucosa peripheral blood same patients. It was found that TIL expressed activation markers CD25 HLA-DR to a higher extent than (p = 0.01), both preferentially CD45RO + phenotype, associated memory cells, in contrast [corrected]. Both contained few natural killer (NK) cells (CD3-CD56+) 0.001), this...
The heterogeneity of the human amyloid proteins SAA and AA was studied. Both could be separated into several fractions by ion‐exchange chromatograpby. Amino acid analysis ion‐exchange‐chromatographed protein showed that main difference in length polypeptide. Thus, it seems original preparation consists a mixture with ranging from 66 to 78 amino residues. By enzymatic degradation three different forms kallikrein, fragments were formed molecular weight very similar thai AA.
Abstract MPB70 is a soluble secreted protein highly expressed in Mycobacterium bovis and strains of bacille Calmette–Guérin (BCG); as such, it candidate for subunit DNA vaccines against tuberculosis. was screened T‐cell epitopes four different inbred mouse strains. Major histocompatibility complex (MHC) H‐2 b ‐expressing mice (C57BL/6) interferon‐γ (IFN‐γ) after stimulation with peptides from the regions 1–20, 41–50, 81–110, 121–150 161–193 sequence. db (B6D2) splenocytes IFN‐γ some same...
Hot volcanic pyroclasts can sinter, vesiculate, and outgas in concert – a combination of processes which remains poorly constrained. And yet this occur coincidently during deposition from pyroclastic density currents, conduit-filling debris, tuffisites. In many these settings, it is the sintering-driven evolution permeability that key to gas transport through evolving deposit. Here, we experimentally theoretically investigate permeable networks sintering hot fragmental systems, are hydrous...
Epidermal cell suspensions from the skin of seven patients with atopic dermatitis (AD) and healthy non‐atopic controls were investigated for presence surface HLA‐DR CD1 antigen, IgE using indirect double‐staining immunofluorescence techniques. Fifty‐seven percent all CDI + 68% cells demonstrated on their surface, indicating that Langerhans (Lc) in AD may be a heterogeneous population regard to characteristics. No found epidermal controls. An attempt lo demonstrate birch pollen antigen Lc...
The terminal complement complex (TCC) was quantified in sera from patients with a genetic deficiency of C8α‐γ or C8β individual contained only trace amounts TCC compared normal serum pool. content increased after mixing the two sera, which consistent reconstitution C8 activity. Only moderate increase obtained zymosan activation whereas mixture resulted high TCC. demonstrated both deficient sera. These results may indicate that functional is present despite deficiency, and pathway function to...

Background
Rituximab (RTX) is effective in treating immune-mediated inflammatory diseases (IMID). Hypogammaglobulinemia may occur under RTX and increase infection risk.
Objectives
Since data are controversial, we evaluated the risk of severe patients with IMID hypogammaglobulinemia therapy.
Methods
We conducted a retrospective single-center study retrieving all treated for IMID. calculated incidence RTX-induced immunoglobulin (Ig) deficiency, type Ig infection,...
