Hematopoietic stem cells (HSCs) are the source of all blood over an individual's lifetime. Diseased HSCs can be replaced with gene-engineered or healthy through HSC transplantation (HSCT). However, current protocols carry major side effects and have limited access. We developed CD117/LNP-messenger RNA (mRNA), a lipid nanoparticle (LNP) that encapsulates mRNA is targeted to cell factor receptor (CD117) on HSCs. Delivery anti-human CD117/LNP-based editing system yielded near-complete...
Abstract Inertial measurement unit (IMU)-based gait analysis systems have become popular in clinical environments because of their low cost and quantitative capability. When a shank is selected as the IMU mounting position, an inverted pendulum model (IPM) can accurately estimate its spatial parameters. However, stride-by-stride estimation parameters using one on each IPMs has not been validated. This study validated parameter method shank-based system. Spatial were estimated via double...
An aerobic, Gram-negative, yellow-pigmented, non-motile bacterium, designated strain KMM 3882 T , was isolated from a marine bivalve ( Anadara broughtoni ) collected Peter the Great Bay, Sea of Japan, and subjected to phenotypic phylogenetic analyses. Strain found exert remarkable inhibitory activity against number Gram-positive micro-organisms. Phylogenetic analysis based on 16S rRNA gene sequences placed within genus Sphingomonas as an independent lineage adjacent dokdonensis DS-4 panni...
Cyclic nucleotide-gated (CNG) ion channels are key mediators underlying signal transduction in retinal and olfactory receptors. Genetic defects CNGA3 CNGB3, encoding two structurally related subunits of cone CNG channels, lead to achromatopsia (ACHM). ACHM is a congenital, autosomal recessive disorder that manifests by photoreceptor dysfunction, severely reduced visual acuity, impaired or complete color blindness photophobia. Here, we report the first canine models for CNGA3-associated...
A Marinomonas-like bacterium, strain KMM 3893T, was isolated from a marine sandy sediment collected close to shore the Sea of Japan and subjected phenotypic phylogenetic study. Comparative 16S rRNA gene sequence analysis confirmed novel strain's assignment genus Marinomonas. Strain 3893T constituted separate phyletic line in Marinomonas, sharing <97 % similarity with respect other recognized Marinomonas species. Chemotaxonomically, contained predominant fatty acids C18 : 1 ω7c, C16 ω7c 0 had...
A Sphingomonas-like bacterium, strain KC7T, was isolated from a marine crustacean specimen obtained the Sea of Japan and subjected to polyphasic study. Comparative 16S rRNA gene sequence analysis positioned novel in genus Sphingomonas as an independent lineage adjacent subclade containing trueperi LMG 2142T, pituitosa EDIVT azotifigens NBRC 15497T. Strain KC7T shared highest similarity (96.1 %) with S. dokdonensis DS-4T 15497T; similarities strains other recognized species were lower...
Abstract Aging increases the risk of abrupt declines in cognitive function after an event that triggers immune system activation (e.g. surgery, infection, or injury). This phenomenon is poorly understood, but rodent models may provide clues. We have previously shown aging (24-mo-old) F344xBN rats generally do not show significant physical impairments. However, their brains mount exaggerated inflammatory response to signals triggered by a peripheral challenge (an intraperitoneal injection...
Ongoing clinical trials for treatment of beta-globinopathies by gene therapy involve the transfer beta-globin gene, which requires integration three to four copies per genome in most target cells. This high proviral load may increase toxicity, potentially limiting safety this and relegating its use total body myeloablation. We hypothesized that introducing an additional hypersensitive site from locus control region, complete sequence second intron ankyrin insulator enhance expression....
Rabies is a zoonotic disease caused by the rabies virus. While salivary glands are important as exit and propagation sites for virus, mechanisms of excretion remain unclear. Here, we investigated histopathology rabid dogs analyzed mechanism into oral cavity. Mandibular parotid 22 three control were used. Mild to moderate non-suppurative sialadenitis was observed in mandibular 19 dogs, characterized loss acinar epithelium infiltration lymphoplasmacytic cells. Viral antigens detected mucous...
Genome Information Broker for Viruses (GIB-V) is a comprehensive virus genome/segment database. We extracted 18 418 complete genomes/segments from the International Nucleotide Sequence Database Collaboration (INSDC, http://www.insdc.org/) by DNA Data Bank of Japan (DDBJ), EMBL and GenBank stored them in our system. The list registered viruses arranged hierarchically according to taxonomy. Keyword searches can be performed data or biological features any GIB-V. GIB-V equipped with BLAST...
Abstract The entire and partial gag regions of human immunodeficiency virus type 1 (HIV‐1) were overproduced in Escherichia coli used for epitope mapping antibodies against p17. We found that a mouse monoclonal antibody to p17, V17 recognizes the mature p17 but not unprocessed Gag proteins containing moiety. Further analysis revealed C‐terminal 12‐amino‐acid region having free C‐terminus. This may be useful monitoring maturation particles.
A simple method for the overproduction in Escherichia coli and purification of major core protein p24 human immunodeficiency virus type 1 (HIV-1) was described. The gag-pol region encoding p24, p15, protease fused to 3' end lacZ gene on plasmid. LacZ-Gag fusion protein, primary product, is designed be cleaved by HIV-1 coexpressed through frameshifting. In fact, its immediate precursor, p25, were produced cells grown at 25C, but not 37C. When gag pol frames in-frame express without...
A large number of complete microorganism genomes has been sequenced and submitted to the public database then incorporated into our genome database, Genome Information Broker (GIB, Author Webpage). However, when comparative genomics is carried out, researchers must be aware that there are protein-coding genes not confirmed by homology or motif search reliable missing. Therefore, we developed a protocol (Gene Trek in Prokaryote Space, GTPS) for finding possible bacterial genomes. GTPS assigns...
In this study, we attempted to classify the methylotrophic yeasts based on diversities of alcohol oxidase (AOD), i.e. zymogram patterns and partial amino acid sequences. According for AOD, members separate into two major lineages, one group involving strains having a single AOD other group, including Pichia methanolica, Candida pignaliae C. sonorensis, showing nine isozymes. Based sequences could be divided five groups, classification agrees mostly with grouping 26S domain D1/D2 rDNA...
Cone cyclic nucleotide-gated channels are tetramers formed by CNGA3 and CNGB3 subunits; subunits function as homotetrameric but exhibits channel only when co-expressed with CNGA3. An aspartatic acid (Asp) to asparagine (Asn) missense mutation at position 262 in the canine (D262N) subunit results loss of cone (daylight blindness), suggesting an important role for this aspartic residue biogenesis and/or function. Asp is located a conserved region second transmembrane segment containing three...
