Results are reported showing that pituitary extracts stimulate growth of 3T3 cells, an established line mouse fibroblast. Cells were cultured in medium containing organic buffer (HEPES; N-2-hydroxyethylpiperazine-N'-2-ethanesulfonic acid), which increases the serum requirement for cell growth. The growth-prompting activity is (a) not dialyzable; (b) thermolabile; (c) protease sensitive; and (d) enhanced by hydrocortisone (a steroid hormone). Activity was quantitatively assayed measurement...

Abstract Cancer homeostasis depends on a balance between activated oncogenic pathways driving tumorigenesis and engagement of stress response programs that counteract the inherent toxicity such aberrant signaling. Although inhibition signaling has been explored extensively, there is increasing evidence overactivation same can also disrupt cancer cause lethality. We show here protein phosphatase 2A (PP2A) hyperactivates multiple engages responses in colon cells. Genetic compound screens...

The adrenocorticotropic hormone (ACTH) inhibits the growth of Y1 mouse adrenocortical tumor cells as well normal in culture but stimulates cell vivo. In this study, we investigated paradoxical effect ACTH on proliferation adrenal and have unmasked a growth-promoting hormone. were arrested G1 phase cycle by serum starvation monitored for progression through S measuring [3H]thymidine incorporation into DNA number nuclei labeled with bromodeoxyuridine. stimulated to progress divide after brief...

Abstract Trypanosomatids are the etiologic agents of various infectious diseases in humans. They diverged early during eukaryotic evolution and have attracted attention as peculiar models for evolutionary comparative studies. Here, we show a meticulous study comparing incorporation detection thymidine analogs BrdU EdU Leishmania amazonensis , Trypanosoma brucei cruzi to monitor their DNA replication. We used BrdU‐ EdU‐incorporated parasites with respective standard approaches: indirect...

We recently reported that paracrine Fibroblast Growth Factor 2 (FGF2) triggers senescence in Ras-driven Y1 and 3T3Ras mouse malignant cell lines. Here, we show although FGF2 activates mitogenic pathways these Ras-dependent cells, it can block proliferation cause a G2/M arrest. These cytostatic effects of are inhibited by PD173074, an FGF receptor (FGFR) inhibitor. To determine which downstream induced FGF2, tested specific inhibitors targeting mitogen-activated protein kinase (MEK),...

Glyceraldehyde 3-phosphate dehydrogenase (GAPDH) is a classical metabolic enzyme involved in energy production and plays role additional nuclear functions, including transcriptional control, recognition of misincorporated nucleotides DNA maintenance telomere structure. Here, we show that the recombinant protein T. cruzi GAPDH (rTcGAPDH) binds single-stranded telomeric DNA. We demonstrate binding to correlates with balance between oxidized reduced forms nicotinamide adenine dinucleotides...

In malignant transformation, cellular stress‐response pathways are dynamically mobilized to counterbalance oncogenic activity, keeping cancer cells viable. Therapeutic disruption of this vulnerable homeostasis might change the outcome many human cancers, particularly those for which no effective therapy is available. Here, we report use fibroblast growth factor 2 ( FGF 2) demonstrate that further mitogenic activation disrupts and strongly sensitizes stress‐targeted therapeutic inhibitors. We...

We have shown that glucocorticoids reversibly change the growth control of rat C6 glioma cells from a transformed to normal pattern. Here we report glucocorticoid hormone hydrocortisone (Hy) modulates structure and function cell surface cytoskeleton. The is cause: (a) increased flattening adhesion solid substrates fibrin layers, (b) inhibition shape triggered by catecholamines cAMP, (c) extensive fibronectin deposition on normally fibronectinless cells' surface, (d) microtubule...

Abstract Fibroblast growth factor 2 (FGF2) is considered to be a bona fide oncogenic factor, although results from our group and others call this into question. Here, we report that exogenous recombinant FGF2 irreversibly inhibits proliferation by inducing senescence in Ras-dependent malignant mouse cells, but not immortalized nontumorigenic cell lines. Cells of the Y-1 corticoadrenal line are: (a) functional, (b) cell cycle-arrested by adrenocorticotropic hormone (ACTH), (c) tumorigenic, and (d) c-Ki-ras overexpressing. We here report that phorbol ester 12-myristate 13-acetate (PMA) mimics all ACTH-specific effects in cells, namely: steroid-ogenesis stimulation, cycle block, shape change. In addition, both ACTH PMA caused a rapid transient induction c-fos proto-oncogene while having no effect on mRNA steady state levels. Dibutyryl cAMP,...
