Significance In vertebrates, the transmembrane protein Smoothened (Smo) accumulates in ciliary membrane when cells receive Hedgehog (Hh) signal. The presence of Smo primary cilia at baseline conditions has been postulated, but not directly observed. We used highly sensitive single-molecule imaging live to track and analyze dynamics individual molecules cilia, only after treatment with pathway agonists also low, levels. both conditions, bind distinct sites bases different dissociation...

The Hedgehog-signaling pathway is an important target in cancer research and regenerative medicine; yet, on the cellular level, many steps are still poorly understood. Extensive studies of bulk behavior key proteins established that during signal transduction they dynamically localize primary cilia, antenna-like solitary organelles present most cells. secreted Hedgehog ligand Sonic (SHH) binds to its receptor Patched1 (PTCH1) causing inactivation delocalization from cilia. At same time,...

Abstract Many bacteria and most archaea possess a crystalline protein surface layer (S-layer), which surrounds their growing topologically complicated outer surface. Constructing macromolecular structure of this scale generally requires localized enzymatic machinery, but regulatory framework for S-layer assembly has not been identified. By labeling, superresolution imaging, tracking the (SLP) from C. crescentus , we show that 2D self-assembly is sufficient to build maintain in living cells...

Super-resolution (SR) microscopy has been used to observe structural details beyond the diffraction limit of ∼250 nm in a variety biological and materials systems. By combining this imaging technique with both computer-vision algorithms topological methods, we reveal quantify nanoscale morphology primary cilium, tiny tubular cellular structure (∼2–6 μm long 200–300 diameter). The cilium mammalian cells protrudes out plasma membrane is important many signaling processes related...

In this study, PET (intrinsic viscosity of 1.05 dl/g) was melt processed with a horizontal isothermal bath (HIB) treatment. Tensile properties fiber samples were highly increased by using the HIB. The process-structure-property relationship control (no HIB) and HIB characterized tensile testing, differential scanning calorimetry, birefringence measurement, electron microscopy hot-air shrinkage measurements. It found that samples, which had been subjected to post-drawing process, high degree...

Abstract Bacteria assemble the cell envelope using localized enzymes to account for growth and division of a topologically complicated surface 1–3 . However, regulatory pathway has not been identified assembly maintenance layer (S-layer), 2D crystalline protein coat surrounding curved 3D variety bacteria 4,5 By specifically labeling, imaging, tracking native purified RsaA, S-layer (SLP) from C. crescentus , we show that self-assembly alone is sufficient maintain in vivo monitoring location...

In this study, xanthan gum and poly (ethylene oxide) (PEO) were blended via hot melt extrusion. Results showed that PEO acted as a plasticiser lowered the viscosity of gum/PEO blends. For example, flow rate torque level observed to decrease at increasing percent compositions in blend. Breakdown testing demonstrated accelerates breakdown rates compared 100 % extrudate samples.
