To determine the effect of infertility-linked sperm phospholipase Cζ (PLCζ) mutations on their ability to trigger oocyte Ca(2+) oscillations and development, also evaluate potential therapeutic utility wild-type, recombinant PLCζ protein for rescuing failed activation embryo development.

The synthesis and biological evaluation of new M(I)(CO)3(NNO) (M = Re, 99mTc) complexes attached to the antitumor agent 2-(4-aminophenyl)benzothiazole are reported. fluorescent rhenium complex enters MCF-7 breast cancer cells but does not enter normal HFFF-2 MRC-5 cells. analogous radioactive 99mTc produces fast blood soft tissue clearance when administered healthy mice. These promising candidates for developing radiopharmaceuticals imaging (99mTc) targeted radiotherapy (186Re, 188Re) cancer.

Calmodulin (CaM) association with the cardiac muscle ryanodine receptor (RyR2) regulates excitation–contraction coupling. Defective CaM–RyR2 interaction is associated heart failure. A novel CaM mutation (CaM F90L ) was recently identified in a family idiopathic ventricular fibrillation (IVF) and early onset sudden death. We report first biochemical characterization of . confers deleterious effect on protein stability. Ca 2+ ‐binding studies reveal reduced affinity loss co‐operativity....

Is it possible to improve clinical visualization of phospholipase C zeta (PLCζ) as a diagnostic marker sperm oocyte activation capacity and male fertility? Poor PLCζ efficacy using current protocols may be due steric or conformational occlusion native PLCζ, hindering antibody access, is significantly enhanced antigen unmasking/retrieval (AUM) protocols. Mammalian mediated via series intracellular calcium (Ca2+) oscillations induced by sperm-specific PLCζ. represents not only potential...

Sperm PLCζ (phospholipase Cζ) is a distinct phosphoinositide-specific PLC isoform that proposed to be the physiological trigger of egg activation and embryo development at mammalian fertilization. Recombinant has ability Ca²⁺ oscillations when expressed in eggs, but it not known how activity regulated sperm or eggs. In present study, we have transfected CHO (Chinese-hamster ovary) cells with fused either YFP (yellow fluorescent protein) luciferase found PLCζ-transfected did display...

Humanin is a 24-mer peptide first reported in the early 2000s as new neuroprotective/cytoprotective factor rescuing neuronal cells from death induced by various Alzheimer's disease-associated insults. Nowadays it known that humanin belongs to novel class of so-called mitochondrial-derived peptides (which are encoded mitochondrial DNA) and has been shown exert beneficial cytoprotective effects series vitro and/or vivo experimental models human diseases, including not only neurodegenerative...
