Bacitracin is a broad-spectrum cyclic peptide antibiotic mainly produced by Bacillus, precursor amino acid supply served as the critical role during its synthesis. In this study, we systematically engineered branch-chain (BCAA) modules for bacitracin production. Firstly, demonstrated that Ile and Leu acted limiting precursors synthesis, BCAA synthetic pathways were strengthened via simultaneous overexpression of, feedback-resistance acetolactate synthase IlvBNfbr, 2-isopropylmalate...

A gene encoding a putative multicopper oxidase (MCO) was cloned from the soil bacterium Klebsiella sp. 601 and its corresponding enzyme overexpressed in an Escherichia coli strain. MCO is composed of 536 amino acids with molecular mass 58.2 kDa. Theoretical calculation gave pI value 6.11. The acid sequence strongly homologous to that E. CueO similarity 90% identity 78%. Unlike CueO, contains extra 20 close C-terminus. purified homogeneity by Ni-affinity chromatography. capable using DMP...

Abstract Background Structural comparison between bacterial CueO and fungal laccases has suggested that a charged residue Glu (E106) in replaces the corresponding Phe at gate of tunnel connecting type II copper to protein surface an extra α-helix (L351-G378) near I site covers substrate binding pocket might compromise electron transfer from copper. To test this hypothesis, several mutants were made Klebsiella sp . 601 multicopper oxidase, which is highly homologous E. coli with similarity...

Abstract Porcine dermal collagen is successfully digested by clostridium histolyticun collagenases (CHC) at varying compositions and conditions. Collagen fragments thus prepared, with significantly low molecular weight but visible denaturation temperatures and/or triple helix structures, may be useful for clinical applications. As evidenced Sodium dodecyl sulfate - polyacrylamide gel electrophoresis (SDS-PAGE) permeation chromatography (GPC) analyses, relatively high weights of α 1 , 2...
