The display of full-length antibody on the cell surface was achieved by fusing a transmembrane domain platelet-derived growth factor receptor (PDGFR) to C-terminus heavy chain constant region. A unique four-way ligation strategy was developed for rapid construction of a full-length antibody library. mammalian expression vector constructed that contained dual cassettes and sequences recognized by the restriction enzymes BsmBI, BstXI, SfiI. Both light-chain variable domain heavy-chain genes were inserted into in one step ligation, bivalent antibodies displayed on cell surfaces. Using this strategy, only 2 weeks required to successfully construct high-quality, human libraries.
