Endolysosomal compartments maintain cellular fitness by clearing dysfunctional organelles and proteins from cells. Modulation of their activity offers therapeutic opportunities. Quantification cargo delivery to and/or accumulation within endolysosomes is instrumental for characterizing lysosome-driven pathways at the molecular level monitoring consequences genetic or environmental modifications. Here we introduce LysoQuant, a deep learning approach segmentation classification fluorescence...

Abstract Intervertebral disc (IVD) degeneration is a spinal disorder that triggers an inflammatory response and subsequent development of pseudoarthrosis. The aim the present study to elucidate role extracellular signal‐regulated kinase (ERK) pathway in inflammation‐induced IVD cells. Inflammatory human nucleus pulposus (NP) cells (NPCs) were induced using tumor necrosis factor‐α ERK was blocked selective molecule‐based inhibitor U0126. Gene expression catabolic anabolic markers,...

Hunter's syndrome (mucopolysaccharidosis type II) is a rare X-linked lysosomal storage disorder caused by mutations in the iduronate-2-sulfatase (IDS) gene. Motivated case of child affected this syndrome, we compared intracellular fate wild-type IDS (IDSWT) and four nonsense (IDSL482X, IDSY452X, IDSR443X, IDSW337X) generating progressively shorter forms associated with mild to severe disease. Our analyses revealed formylation all at cysteine 84, which prerequisite for enzymatic activity....

Abstract Background: Intervertebral disc (IVD) degeneration is a spinal disease caused by trauma and/or repetitive mechanical overloading of the spine which triggers inflammatory response pathways. Long-term inflammation may lead to development pseudoarthrosis. The aim present study was elucidate role extracellular signal-regulated kinase (ERK) pathway in inflammation-induced IVD cells. Methods: Inflammatory human nucleus pulposus cells (NPC) were stimulated using tumor necrosis factor alpha...
