The reduction in podocyte density to levels below a threshold value drives glomerulosclerosis and progression ESRD. However, technical demands prohibit high-throughput application of conventional morphometry for estimating density. We evaluated method using single paraffin-embedded formalin-fixed sections. Podocyte nuclei were imaged indirect immunofluorescence detection antibodies against Wilms' tumor-1 or transducin-like enhancer split 4. To account the large size relation section...

DNA vaccines encoding for murine rotavirus proteins VP4, VP6, or VP7 were tested in adult BALB/cmice their ability to induce immune responses and protect against challenge. A gene gun was used inoculate into the epidermis. Rotavirus-specific serum antibodies, as measured by ELISA, virus-specific cytotoxicT lymphocyte generated each of three vaccines, but virus-neutralizing antibodies detected only mice that with VP4 VP7. Efficacy determined challenge 100 ID50 homotypic rotavirus. Each...

Three weeks after myocardial infarction (MI) in the rat, remodeled hypertrophy of noninfarcted myocardium is at its maximum and heart a compensated stage with no evidence failure. Our hemodynamic measurements this showed slight but insignificant decrease +dP/dt significantly higher left ventricular end-diastolic pressure. To investigate basis diastolic dysfunction, we explored possible defects beta-adrenergic receptor-G(s/i) protein-adenylyl cyclase-cAMP-protein kinase A-phosphatase pathway,...

Norwalk virus, an important cause of epidemic, acute, nonbacterial gastroenteritis in adults and children, has eluded adaptation to tissue culture, the development animal model, molecular cloning. In this study, a portion viral genome encoding immunoreactive region was cloned from very small quantities infected stool using sequence-independent single primer amplification. Six overlapping complementary DNA (cDNA) clones were isolated by immunologic screening. The expressed recombinant protein...

Hyperinsulinemia is commonly viewed as a compensatory response to insulin resistance, yet studies have demonstrated that chronically elevated may also drive resistance. The molecular mechanisms underpinning this potentially cyclic process remain poorly defined, especially on transcriptome-wide level. Transcriptomic meta-analysis in >450 human samples fasting reliably and negatively correlated with INSR mRNA skeletal muscle. To establish causality study the direct effects of prolonged...

Fibroblast growth factor 1 (FGF1) has been recently characterized as a potent insulin sensitizer that regulates adipose tissue remodeling, but the physiological role of FGF1 remains unclear. This study measured serum levels for first time in patients with newly diagnosed type 2 diabetes mellitus (T2DM), and further explored correlations between various metabolic parameters T2DM. Serum were determined using ELISA age-, sex- BMI- matched subjects normal glucose tolerance (NGT) (n=80) T2DM...
