The in vitro metabolism of radioactively labeled androstenedione and testosterone three different areas the canine central nervous system has been studied. Incubation 4-14C with minced preparations dog hypothalamus, pituitary, limbic resulted formation 14C-labeled 5α-androstane-3,17-dione, testosterone, 17β-hydroxy-5α-androstane-3-one. When testosterone-7-3H was used as substrate, 3H-labeled 17β-hydroxy-5α-androstane-3-one, androstenedione, 5α-androstane-3,17-dione were noted. These...
The Student Partnerships for Advanced Research and Knowledge (SPARK) program provides high school students their science educators the opportunity to become visiting researchers Brookhaven National Laboratory’s scientific facilities— just like thousands of use them research each year. Participants in this year-round conduct hypothesis-driven through collaborations with experts at Synchrotron Light Source II. This experience enables actively contribute community, obtain a real experience,...
ABSTRACT Studies were conducted in castrated golden hamsters to assess whether sexual dimorphism and sensitivity sex steroid hormones the rodent Harderian gland are mediated by an interaction of androgens with specific intracellular receptors. Physical properties, binding kinetics stereospecificity androgen receptor analysed using [ 3 H]mibolerone as radioligand. The presence H]mibolerone–androgen complexes a sedimentation coefficient 7–8S was demonstrated cytosol linear sucrose gradient...
We examined herpes simplex virus (HSV)-infected human HEp-2 cells or porcine that express entry mediator (HVEM) for and receptor protein interactions. Antibody to HVEM, its viral ligand gD, coimmunoprecipitated several similar proteins. A prominent 110-kDa coprecipitated was identified as gH. The HVEM/gD/gH complex detected with mild stringent cell lysis conditions. It did not form in infected HSV-1(KOS)Rid1 null lacking gH, gL. Thus, a forms through physical associations of at least
We isolated a human cDNA by expression cloning and characterized its gene product as new protein that enables entry infection of herpes simplex virus (HSV). The gene, designated hfl-B5, encodes type II cell surface membrane protein, B5, is broadly expressed in primary tissue lines. It contains high-scoring heptad repeat at the extracellular C terminus predicted to form an alpha-helix for coiled coils like those cellular SNAREs or some viral fusion proteins. A synthetic 30-mer peptide has...
Herpes simplex virus (HSV) enters and infects most cultured cells. We have found that swine testis cells (ST) produce yields of infectious HSV-1 up to four orders magnitude lower than those human embryonic lung (HEL) HEp-2 because a defect in entry. For ST cells, binding is reduced, DNA from input cannot be detected, proteins are not synthesized. Polyethylene glycol treatment after exposure HSV allows viral entry, protein synthesis, productive infection. Transfection genomic bypasses the...
An equal dose (15, μCi) of high specific activity [3H]testosterone, [3H]androstenedione and [3H]5α-dihydrotestosterone was administered intravenously to 3 groups 8 castrated male rats. One hour later the concentration radioactivity determined in pituitary gland, hypothalamus, preoptic region, mesencephalic tegmentum, hippocampus, amygdala, parietal cortex, plasma. In all cases highest uptake observed pituitary, pituitary/cortex ratio being 13.1 for dihydrotestosterone, 5.6 testosterone, 2.9...
Seaweeds are ecosystem engineers that can serve as habitat, sequester carbon, buffer ecosystems against acidification, and, in an aquaculture setting, represent important food source. One health issue regarding the consumption of seaweeds and specifically, kelp, is accumulation some trace elements concern within tissues. As atmospheric CO
Micrometeorites (MMs) are small particles that account for most of the extraterrestrial material deposited on Earth. Synchrotron X-ray fluorescence and diffraction allowed chemical mineral characterization to distinguish MM from atmospheric particulate. The relative components iron, nickel, other elements were considered in identification ferrous while high amounts titanium an indication origin. Out 100 samples collected by school students teachers, eight taken a synchrotron analysis. Of...
A technique is described for generating large well diffracting crystals from conditions that yield microcrystals. Crystallization using this both rapid (crystals appear in <1 h) and robust (48 out of 48 co-crystallized with a fragment library, compared 26 conventional hanging drop). Agarose gel used to exclude nucleation inducing elements the remaining crystallization cocktail. The chemicals cocktail are partitioned into high concentration components (presumed induce aggregation by...

14683 Background: Steroid sulfatase (STS) is a new target for the treatment of steroid hormone-dependent diseases such as breast, prostate or endometrial cancer. In breast cancer, estrogens play major role in establishment disease and between one to two- thirds tumours are estrogen receptor (ER) positive. Despite current hormonal treatments, improvement still necessary achieve better control & improve outcome. BN83495 non-steroidal, non-estrogenic, potent, irreversible STS inhibitor that...
