General anesthetics induce loss of consciousness, a global change in behavior. However, corresponding activity the context defined cortical cell types has not been identified. Here, we show that spontaneous mouse layer 5 pyramidal neurons, but no other type, becomes consistently synchronized vivo by different general anesthetics. This heightened neuronal synchrony is aperiodic, present across large distances, and absent neurons presynaptic to neurons. During transition from anesthesia,...

Cortical circuits are composed predominantly of pyramidal-to-pyramidal neuron connections, yet their assembly during embryonic development is not well understood. We show that mouse Rbp4-Cre cortical neurons, transcriptomically closest to layer 5 pyramidal display two phases circuit in vivo. At E14.5, they form a multi-layered motif, only near-projecting-type neurons. By E17.5, this transitions second motif involving all three types, analogous the adult types. In vivo patch clamp recordings...

Schwann cells elaborate myelin sheaths around axons by spirally wrapping and compacting their plasma membranes. Although actin remodeling plays a crucial role in this process, the effectors that modulate cell cytoskeleton are poorly defined. Here, we show cytoskeletal regulator, neural Wiskott-Aldrich syndrome protein (N-WASp), is upregulated myelinating coincident with elaboration. When N-WASp conditionally deleted at onset of myelination, continue to ensheath but fail extend processes...

Summary Targeting genes to specific neuronal or glial cell types is valuable both for understanding and repairing brain circuits. Adeno-associated viral vectors (AAVs) are frequently used gene delivery, but targeting expression a challenge. We created library of 230 AAVs, each with different synthetic promoter designed using four independent strategies. show that ~11% these AAVs specifically target in the mouse retina, brain, non-human primate retina vivo , human vitro . demonstrate...

Neurons must faithfully encode signals that can vary over many orders of magnitude despite having only limited dynamic ranges. For a correlated signal, this range constraint be relieved by subtracting away components the signal predicted from past, strategy known as predictive coding, relies on learning input statistics. However, statistics natural also very short time scales e.g., following saccades across visual scene. To maintain reduced transmission cost to with rapidly varying...

Quantum conditions on the control of dynamics a system coupled to an environment are obtained. Specifically, consider initially in subspace H0 dimensionality M0, which evolves populate subspaces H1, H2 dimensionalities M1, M2. Then, there always exists initial state that does not evolve into if M0>dM2, where 2⩽d⩽(M0+M1+M2)2 is number operators Kraus representation. Note, significantly, maximum d can be far smaller than dimension bath. If this condition satisfied, then from avoids only...

Summary Cortical circuits are composed predominantly of pyramidal-to-pyramidal neuron connections, yet their assembly during embryonic development is not well understood. We show that layer 5 pyramidal neurons, identified through single cell transcriptomics, display two phases circuit in vivo. At E14.5, a multi-layered motif, type, forms. This motif transient, switching to second involving all three types, by E17.5. In vivo targeted recordings and two-photon calcium imaging neurons reveal...
