Capillary electrophoresis (CE) is a powerful separation technique offering quick and efficient analyses in various fields of bioanalytical chemistry. It characterized by many well‐known advantages, but one, which perhaps the most important for this application field, somewhat overlooked. possibility to perform chemical biochemical reactions at nL scale inside capillary. There are two basic formats applicable purpose, heterogeneous homogeneous. In former, one reactant immobilized onto...
Diamine oxidase was prepared from pea (Pisum sativum) seedlings by a new purification procedure involving two h.p.l.c. steps. We studied the optical and electrochemical properties of homogeneous enzyme also analysed hydrolysed protein several methods. The data presented here suggest that carbonyl cofactor diamine is firmly bound pyrroloquinoline quinone.
The binding ability of a drug to plasma proteins influences the pharmacokinetics drug. As result, it is very important issue in new development. In this study, affinity capillary electrophoresis, electrophoresis with frontal analysis, and Hummel Dreyer methods internal external calibration were used study between bovine serum albumin salicylic acid. constant was measured by all these approaches including equilibrium dialysis, which considered be reference method. comparison results other...
Pharmacokinetic and pharmacodynamic properties of a chiral drug can significantly differ between application the racemate single enantiomers. During development, characteristics candidate compounds have to be assessed prior clinical testing. Since biotransformation influences actions in an organism, metabolism studies represent crucial part such tests. Hence, optimized economical capillary electrophoretic method for on‐line enantioselective mediated by cytochrome P450 enzymes was developed....
A new method for the determination of kinetic and inhibition parameters cytochromes P450 reactions by means on-line CE was developed. It is based on transverse diffusion laminar flow profiles methodology introduced Krylov et al. that injection procedure modified. The solutions an enzyme its substrates are injected hydrodynamic pressure as a series repeated consecutive plugs. Proposed three plugs surrounded with represents certain trade-off to obtain reaction mixture satisfying homogeneity...
