A G-quadruplex-specific DNA endonuclease was constructed by fusing a RHAU G-quadruplex recognition domain with Fok1 cleavage domain, providing useful tool for detection of structures.
Abstract Tyrosine kinase 2 (TYK2) is a member of the JAK family nonreceptor tyrosine kinase, together with JAK1, JAK2, and JAK3. JAKs are important signaling mediators many proinflammatory cytokines represent compelling pharmacological targets for autoimmune inflammatory diseases. Pan-acting small-molecule inhibitors were approved treatment rheumatoid arthritis ulcerative colitis. However, their limited selectivity among members have led to undesirable side effects, driving search toward...
G-quadruplexes (G4s) are reported to present on the SARS-CoV-2 RNA genome and control various viral activities. Specific ligands targeting those nucleic acid structures could be investigated as promising detection methods or antiviral reagents suppress this menacing virus. Herein, we demonstrate binding between a G4 structure in of fluorescent probe created by fusing parallel-G4 specific RHAU53 cyan protein. The RHAU peptide was easily detected under fluorescence spectrometer. drawbacks...
