Abstract Limbic-predominant age-related TDP-43 encephalopathy neuropathologic change (LATE-NC), is common in elderly brains and often seen conjunction with Alzheimer’s disease (ADNC). LATE-NC typically begins the amygdala spreads to hippocampus neocortex. Whether it contributes hippocampal atrophy Down syndrome (DS) remains unexplored. We analyzed volumes neuropathological burden 12 DS cases 54 non-DS AD related neurodegenerative pathologies (ADRNP) using 7 Tesla (7T) postmortem ex vivo MRI....
Numerous research groups worldwide have focused on postmortem imaging to bridge the resolution gap between clinical neuroimaging and neuropathology data. We developed a standardized protocol for brain embedding, imaging, processing, facilitating alignment antemortem MRI, pathology observe atrophy structural damage progression over time. Using 7T ex vivo we explore potential correlation of amygdala hippocampal with neuropathological burden in both Down syndrome (DS) Alzheimer's disease (AD)...
In this work, we present an image alignment pipeline that facilitates ongoing Alzheimer’s disease (AD) white matter pathology studies. Following our recently developed ex-vivo 7T cutting-guide container [1], in demonstrate a standardized (between histopathology and MRI) procedure significantly reduces human effort eliminate the need for manual alignment.
Abstract Purpose To develop an RF shimming approach for operating the 2 nd Generation Tic Tac Toe Coil System (60 transmit channels integrated with 32-channel receive insert) routine use in 7T neuro MRI on single mode. Methods simulations were performed and used to non-subject-specific shim cases over three anatomically detailed head models: adult male, female, child female. Multi-ROI strategies developed implemented. B 1 + maps in-vivo images acquired mode of a scanner using derived from...
Abstract Background As the usage of MRI for biomedical research increases, an ongoing goal extending usefulness MRIs has been increase magnet’s field strength to 3T, 7T, and beyond. The 7T recently given FDA‐approval clinical use, due recognition its safety ability characterize “smaller structures subtle pathologies” 1 when compared 3T (Fig. 1.) Technological developments at 7 Tesla Bioengineering Research Program (7TBRP) University Pittsburgh is currently enabling neuroimaging translational...
Abstract Background Employing a custom 60‐channel single‐transmit/32‐channel receive Tic‐Tac‐Toe (TTT) coil in 7T MRI imaging has enhanced the resolution and contrast for identifying White Matter Hyperintensities (WMHs), which are critical markers Alzheimer’s disease related dementias (AD/ADRD). Method The method involves resecting left hemisphere of brain, excluding cerebellum, followed by embalming 10% formalin. 1.5% agar 30% sucrose weight heated mixed embedding container. brain is...
Abstract Background High‐resolution and high‐contrast postmortem MRI can unveil structural abnormalities overlooked by antemortem MRI, making it well‐suited for targeted pathology assessments. In community cohorts, limbic‐predominant age‐related TDP‐43 encephalopathy (LATE) is linked with smaller amygdala [Makkinejad 2019 Neurobiol Aging] hippocampal volumes [Josephs 2015 Ann Neurol]. Whether a similar association exists in Down syndrome (DS) remains unexplored. This study investigates the...

A 3D printed re-usable box enclosure and two-piece cutting guide were developed to produce high quality 7T MR images that can be easily registered gross anatomy histology. Cuts made parallel or at multiple angles 5mm intervals in both the axial coronal orientations.
We present a comprehensive ex-vivo brain ultra-high field MRI workflow that addresses transmit magnetic in-homogeneities without the need to build dedicated coil. Additionally, we show 7T T1w with high quality grey white matter contrast is feasible MP2RAGE .
In this work, we computationally and experimentally evaluated the B1+ field SAR generated by a 60-channel Tic-Tac-Toc RF coil at 7T MRI scanner on variety of participants, head models positions. On three anatomically detailed human models, found slight changes in intensity distribution over whole brain when was moved out from top coil. This confirmed both simulation experimental maps. Our results highlighted impact imaging setup quality.
A 60-channel transmit array for 7T neuro MRI was implemented using the Tic-Tac-Toe coil design. The and receive insert are open in front to promote patient comfort reduce claustrophobia. By performing RF shimming on three head models, we optimized a range of ages sizes/shapes use sTx mode. Simulations, experimental B1+ maps in-vivo images demonstrate consistency across subjects, showing extended coverage into temporal lobe, cerebellum, brainstem C5-C6 spinal cord segments. High quality...
The recently developed 60-channel Tic-Tac-Toe transmit coil was analyzed for use in either single or parallel systems at 7T. With the TTT coil, excellent homogeneity can be achieved and safe levels of SAR provided regardless which system (sTx pTx) is used.
The ability to continuously prototype and 3D print new containers as well using an improved transmit coil has led major improvements in ex-vivo image quality. This also allowed for a more automated registration process of the MR images gross anatomy
We investigated the impact of coupling (magnitude and phase) on B 1 + field distribution intensity a 30-channel double-rowed Tic-Tac-Toe (TTT) RF array. found that by changing TTT coil modules, we are able to modify from single channel how is spread between different Z levels This effect was seen in both simulations experimental maps.
Abstract Background White matter hyperintensities (WMHs) are neuroimaging features associated with Alzheimer’s disease (AD). Ex‐vivo studies may enable better characterization of WMHs pathological substrates. In this work, we present a novel method that leverages in‐vivo and ex‐vivo MRIs histopathology to analyze the progression in AD. Method A single subject’s MRI scans at four different time points were analyzed. brain left hemisphere done using 16‐Tx 32‐Rx TicTacToe coil 3 3‐D printed...
