Metal‐Organic Framework Encapsulation Preserves the Bioactivity of Protein Therapeutics

ABSTRACT
Protein therapeutics are prone to lose their structure and bioactivity under various environmental stressors. This study reports a facile approach using nanoporous material, zeolitic imidazolate framework-8 (ZIF-8), as an encapsulant for preserving the prototypic protein therapeutic, insulin, against different harsh conditions that may be encountered during storage, formulation, transport, including elevated temperatures, mechanical agitation, organic solvent. Both immunoassay spectroscopy analyses demonstrate preserved chemical stability structural integrity of insulin offered by ZIF-8 encapsulation. Biological activity ZIF-8-preserved after storage accelerated degradation (i.e., 40 °C) is evaluated in vivo diabetic mouse model, shows comparable refrigeration-stored (-20 °C). It also demonstrated has low cytotoxicity vitro does not cause side effects vivo. Furthermore, residue can completely removed simple purification step before administration. biopreservation potentially applicable diverse therapeutics, thus extending benefits advanced biologics resource-limited settings underserved populations/regions.
