ABSTRACT
Abstract Current scaffolds applied for bone tissue engineering are still lacking sufficient osteogenic capacity to induce efficient regeneration. Biodegradable microsphere‐type designed achieve the dual‐controlled release of a Chinese medicine (i.e., icariin, ICA) and bioactive ion Mg 2+ ), in order their synergistic effect on inducing osteogenesis. The hydrophobic icariin is preloaded onto MgO/MgCO 3 (1:1 weight ratio) particles at different amounts then encapsulated into biodegradable poly(lactide‐ co ‐glycolide) (PLGA) microspheres (PMI) fixed fraction (20 wt%). Continuous releases from detected, showing dependence amounts. At an optimized moderate loading amount, resulting PMI‐M display strongest activation cell biological behaviors among all designs. By implanting rat calvarial defects 16 weeks, it found that they can effectively enhance new formation, presenting significantly higher osteogenesis than PMg (containing but without icariin) blank PLGA microspheres. Clearly, released ions beneficial osteogenesis, coincorporation exerts supplemental effects which suggest promising strategy regenerate severe injuries by designing dual‐release system.
