Photo‐Facilitated Nitric Oxide‐Triggered Turn‐on Photodynamic Therapy for Precise Antitumor Application

ABSTRACT
Abstract Photodynamic therapy (PDT) is a powerful strategy for tumor with noninvasiveness and desirable efficacy. However, the phototoxicity of photosensitizer after post‐PDT major obstacle limiting clinic applications. Herein, nitric oxide (NO)‐activatable reported turn‐on PDT behavior endoplasmic reticulum (ER) targeting ability precise therapy. Four o ‐thiophenediamine derivatives reaction‐tunable donor/acceptor push‐pull electronic effect are established, systematic structure property relationship observation reveals following features: 1) reactivity against NO can be improved by enhancing electron density further facilitated upon photo‐irradiation. 2) enables intramolecular charge transfer process evoking photosensitizing effect. 3) only derivative ER enrichment exhibited cancer cell ablation compared to photosensitizers localized in lysosome lipid droplet. Thus, efficient inhibition cells both vitro vivo realized based on photo‐controlled strategy. This work provides more insights into developing promising activatable advanced microenvironment trigger.
