ABSTRACT
Abstract N6‐methyladenosine (m 6 A) is rapidly being studied and uncovered to play a significant role in various biological processes as well RNA fate functions, while the effects of defined m A sites are rarely characterized for lack convenient tools. To provide an applicable method remove modification at specific loci, editing system called “ t argeted R N de methylation by S unTag (TRADES)” engineered. In this system, targeting element dCas13b fused ten copies GCN4 peptides which can recruit multiple scFv‐fusion demethylase demethylate target site. Owing design, TRADES more flexible indistinct its wide window. By site‐specific demethylation messenger (mRNA) SON A2699, lifetime successfully prolonged HeLa cells. Meanwhile, negligibly influences other non‐targeted transcripts. also regulate gene expression transcript A‐dependent manner. Moreover, interference occuring HBV HIV replications demonstrates that holds potential viral life cycle regulation clinical applications.
