ABSTRACT
Abstract Minimally invasive thermal therapies have been attempted in the treatment of breast cancer, and immune response induced by these has not fully reported. A clinical trial is performed to determine effect microwave ablation (MWA) early‐stage cancer. The authors perform single‐cell RNA sequencing on peripheral blood mononuclear cells (PBMCs) from six patients before after ablation. NK CD8 + T are activated MWA with increased inhibitory signature but dysfunctional. Enhanced co‐stimulatory CD4 observed frequency ICOS confirmed flow cytometric analysis. After ablation, T‐cell clones expand receptor diversities. Activated antigen receptor‐mediated signaling pathways found B cells. interactions between found, indicating that important antigen‐presenting initiate MWA‐induced response. Blockade CTLA‐4 or PD‐1 post‐MWA PBMCs show higher activity than pre‐MWA PBMCs. This study provide global characteristics systemic pave a way for identification potential targets improve
