Site‐Specific Antibody Assembly on Nanoparticles via a Versatile Coating Method for Improved Cell Targeting

ABSTRACT
Abstract Antibody‐nanoparticle conjugates are promising candidates for precision medicine. However, developing a controllable method conjugating antibodies to nanoparticles without compromising the antibody activity represents critical challenge. Here, facile and generalizable film‐coating is presented using zeolitic imidazole framework‐8 (ZIF‐8) immobilize on various in favorable orientation enhanced cell targeting. Different model therapeutic (e.g., Herceptin) assembled via biomineralized exhibited high loading targeting efficiencies. Importantly, selectively bind ZIF‐8 their Fc regions, which favorably exposes functional Fab regions biological target, thus improving ability of antibody‐coated nanoparticles. In combination, molecular dynamics simulations experimental studies immobilization, efficiency, biofunctionality collectively demonstrate that this versatile site‐specific conjugation provides effective control over leads improved variety
