ABSTRACT
Abstract Human amniotic epithelial cells (hAECs) have shown excellent efficacy in clinical research and prospective applications the treatment of many diseases. However, properties hAECs their proliferative mechanisms remain unclear. Here, single‐cell RNA sequencing (scRNA‐seq) is performed on obtained from tissues at different gestational ages passages during vitro culture. The results showed that proliferation associated with epithelial‐mesenchymal plasticity (EMP) amniogenesis. Freshly isolated, full‐term are identified as mature cells. Once cultured vitro, they observed to rapidly undergo transition (EMT) enter a partial (pEMT) state regain EMP capacities. With continuous development EMT, eventually senescent state. addition SB431542 microcarrier screening enabled effective 3D expansion by 50 fold while maintaining status for further proliferation. This study not only elucidated central mechanism but also optimized culture system so it sufficient generate patients single donor membrane.
