ABSTRACT
Abstract Background Plasma markers of amyloid, tau, and neurodegeneration (ATN) are promising as correlates AD biological clinical features, but data from older African Americans, on change during non‐pharmacological interventions, is limited. Method The Program for American Cognition Exercise (PAACE) randomized cognitively normal adults into a 12‐week successful aging group (SAG) to attend educational sessions or physical activity (PAG) complete supervised in‐home activity. Repeatable Battery the Assessment Neuropsychological Status (RBANS) cardiometabolic risks were assessed pre‐ post‐intervention. Quanterix Simoa® Neuro 4‐Plex E pTau‐181 Advantage Kits used measure plasma concentrations amyloid beta (ABeta) 1‐40 1‐42, glial fibrillary acidic protein (GFAP), neurofilament light (NfL), tau phosphorylated at threonine 181 (pTau‐181). Linear mixed effects regression models modifiers in RBANS measures. Result A set 54 PAACE completers aged 69.3 +/‐ 3.4 years (40 female, 14 male) provided non‐outlier measurements. ABeta decreased ( p =.027), global cognition, visuospatial, delayed memory scores increased, over intervention (maximum value: .040). ATN biomarkers did not differ between SAG PAG baseline, have greater declines than =.003, Figures 1‐2), women had men = .045, Figure 3). Greater age was associated with 1‐40, GFAP, NFL .008). diastolic blood pressure lesser .021) .041). NfL, pTau‐181, suggestive burden neurodegeneration, language score 0495). immediate .035). modify pre‐to‐post‐intervention any summary (minimum =.10). Conclusion Among course nonpharmacological differed by study gender. cognitive function an intuitive fashion.
