Enabling Photo‐Crosslinking and Photo‐Sensitizing Properties for Synthetic Fluorescent Protein Chromophores

ABSTRACT
Abstract Synthetic fluorescent protein chromophores have been reported for their singlet state fluorescence properties and applications in bioimaging, but rarely the triplet chemistries. Herein, we enabled photo‐sensitizing photo‐crosslinking through rational modulations. Extension of molecular conjugation introduction heavy atoms promoted generation reactive oxygen species. Unlike other photosensitizers, these selectively photo‐crosslinked aggregated proteins uncovered interactome profiles. We also exemplified general chromophore‐assisted light inactivation, photodynamic therapy photo induced polymerization. Theoretical calculation, pathway analysis transient absorption spectroscopy provided mechanistic insights this chemistry. Overall, work expands function application synthetic by enabling excited properties.
