Untargeted GC‐MS Metabolomics Reveals Changes in the Metabolite Dynamics of Industrial Scale Batch Fermentations of Streptoccoccus thermophilus Broth

ABSTRACT
An industrial scale biomass production using batch or fed‐batch fermentations usually optimized by selection of bacterial strains, tuning fermentation media, feeding strategy, and temperature. However, in‐depth investigation the metabolome during may reveal new knowledge for better optimization. In this study, first time, authors investigated seven batches performed on five Streptoccoccus thermophilus strains at Chr. Hansen (Denmark) in a real life large process. The study is designed to investigate effects fermentation, line, yeast extract brands untargeted GC‐MS metabolomics. Processing raw data PARAFAC2 revealed total 90 metabolites out which 64 are identified. Partitioning variance according experimental design was ASCA that time their interaction term were most significant effects. brand had smaller impact metabolome, while line showed no effect. This shows metabolic analysis broth provides tool advanced optimization high‐volume‐low‐cost lowering cost, increase yield, augment product quality.
