Efficient Generation of Viral and Integration‐Free Human Induced Pluripotent Stem Cell‐Derived Oligodendrocytes

ABSTRACT
Here we document three highly reproducible protocols: (1) a culture system for the derivation of human oligodendrocytes (OLs) from induced pluripotent stem cells (hiPS) and their further maturation-our protocol generates viral- integration-free OLs that efficiently commit move forward in OL lineage, recapitulating all steps known to occur during vivo development; (2) method isolation, propagation maintenance neural (NSCs); (3) production, perinatal rodent brain-derived NSCs. Our unique systems rely on series chemically defined media, specifically designed carefully characterized each developmental stage as they advance progenitors mature, myelinating cells. We are confident these protocols bring our field step closer efficient autologous cell replacement therapies disease modeling. © 2016 by John Wiley & Sons, Inc.
