ABSTRACT
Zinc finger E-box binding homeobox 1 (ZEB1) is a transcription factor that plays central role in the epithelial to mesenchymal transition (EMT) of cancer cell lines. Studies on its regulation have mostly focused negative 3'UTR miR200c. Interestingly, it has been previously reported androgen receptor (AR) regulates ZEB1 expression breast and prostate cancers. In order validate this, various promoter deletions were cloned into luciferase reporter system elucidate contribution two putative response elements (AREs). The vivo AR was also assessed lines after R1881 treatment using qPCR with specific antigen (PSA) as positive control. We discovered upregulates levels 10-fold only contains distal ARE. However, when proximal ARE included, no additional activation apparent or hormone independent variant, AR-V7. Furthermore, we demonstrate here construct containing both AREs activates even AR-null DU145 PC3. Incubation AR-positive line, LNCaP R1881, failed substantially increase ZEB1. Despite presence region, appears can be induced without AR. addition, region around potent repressor
