Mesenchymal stem cells administered after liver transplantation prevent acute graft-versus-host disease in rats

ABSTRACT
Acute graft-versus-host disease is a serious and life-threatening complication of liver transplantation (LT) that occurs in 1% to 2% allograft recipients. It associated with high mortality rate, effective therapies are lacking. In our established rat model, relative decrease regulatory T cells (Tregs) was previously shown be acute after (LT-aGVHD). Mesenchymal stem (MSCs) have been used treat allogeneic hematopoietic cell transplantation, they induce Tregs, which immunomodulatory effects. this study, when treatment donor- or recipient-derived MSCs administered from day 8 14 the typical symptoms LT-aGVHD started, recipients were not cured, their survival time prolonged. However, different origins 0 6 LT, survived significantly longer than control group, surviving MSC-treated rats did show symptoms. vivo tracings carboxyfluorescein diacetate succinimidyl ester-stained significant accumulations target organs administration. Flow cytometry analysis showed Treg ratios peripheral blood more higher for groups versus group. More immunohistochemically stained forkhead box P3-positive also found intestines Further investigations function could increase ratio mixed lymphocyte reaction (MLR) lead greater reduction MLR proliferation vitro. conclusion, post-LT administration either donor recipient origin prevent onset model.
