Neuroprotective effects of 20(S)‐protopanaxatriol (PPT) on scopolamine‐induced cognitive deficits in mice

ABSTRACT
20(S)-protopanaxatriol (PPT), one of the ginsenosides from Panax ginseng, has been reported to have neuroprotective effects and improve memory. The present study was designed investigate protective effect PPT on scopolamine-induced cognitive deficits in mice. Male Institute Cancer Research mice were pretreated with 2 different doses (20 40 μmol/kg) for 27 days by intraperitoneal injection, scopolamine (0.75 mg/kg) injected intraperitoneally 9 induce memory impairment. Thirty minutes after last pretreatment, locomotor activity firstly examined evaluate motor function Then, memory-related behaviors evaluated, related mechanism further researched. It founded that treatment significantly reversed impairment object location recognition experiment, Morris water maze test, passive avoidance task, showing memory-improving effects. also improved cholinergic system reactivity suppressed oxidative stress, indicated inhibition acetylcholinesterase activity, elevation acetylcholine levels, increasing superoxide dismutase lowering levels malondialdehyde hippocampus. In addition, expression Egr-1, c-Jun, cAMP responsive element binding hippocampus elevated administration. These results suggest may be a potential drug candidate deficit Alzheimer's disease.
