MicroRNA-200C and -150 play an important role in endothelial cell differentiation and vasculogenesis by targeting transcription repressor ZEB1

ABSTRACT
To investigate the role of miRNA in controlling human embryonic stem (hES) cell differentiation toward endothelial lineage and chick blood vessel formation, undifferentiated hES cells were first cultured on Matrigel-coated flasks growth medium-2 (EGM-2) to initiate (EC) differentiation. CD146(+) isolated from differentiating expanded vitro. The vitro positive for EC markers, capable Ac-LDL uptake, lectin binding, formation vascular structures vivo. gain/loss-of-function analyses revealed that miR-150 miR-200c crucial Transcriptional repressor zinc finger E-box-binding homeobox 1 (ZEB1) was identified as communal target gene miRNA-200C -150, inhibition ZEB1 required or -150 mediated expressions. Moreover, we demonstrated could transcriptionally repress expression through direct binding promoters genes. Finally, also miRNA-200c played an important by vivo developing embryos, blocking signaling CD146-positive rescue inhibitory effects inhibiton vasculogenesis. Our findings play specification vasculogenesis targeting ZEB1.
