ABSTRACT
PURPOSE: A variety of adjuvant treatments and cytoprotective agents have been proposed to lessen the toxicity radiation therapy. The following study was designed evaluate benefit six or combinations using anastomotic bursting strength as a measure transmural injury. METHODS: 40-Gy consisted following. Seventy-two male Sprague-Dawley rats were divided into eight equal groups: nonradiated control, radiated untreated treated groups. radioprotective included ribose-cysteine (RibCys), WR-2721, glutamine, vitamin E, MgCl2/adenosine triphosphate, RibCys/glutamine in combination. Radiated animals received 40 Gy abdomen. Two weeks after radiation, all underwent small bowel colonic resection with primary anastomosis. Animals sacrificed one week postoperatively, at which time anastomoses evaluated strengths determined. 70-Gy same protocol repeated for five groups nine nonradiated, untreated, three receiving RibCys (8 mmol/kg), (20 WR-2721. All doses. RESULTS: In group, there 10 radiation-related deaths 6 leaks among 70 studied. None differences between significant. Nonradiated control group large pressures significantly elevated compared Compared controls, significant improvements RibCys, RibCys-glutamine, E improvement survived. died, four WR-2721 died (P=0.03), mmol/kg) survived (P=0.08). CONCLUSIONS: gave consistent protection against lower incidence treatment-related potentially greater effects may make more clinically useful than
