ABSTRACT
The epidermal growth factor receptor (EGFR) is expressed at different levels in many cell types and found overexpressed cancers. EGFR expression increased or decreased response to extracellular stimuli. We examined the effect of c-Jun on promoter activity. used DNAse I foot-printing analysis determine binding activator protein 1 (AP-1) region. also cotransfection experiments western blotting AP-1 family members expression. was able bind least seven sites Cotransfection MCF-7 cells with a vector reporter resulted 7-fold increase JunB, but not c-fos, enhanced An A-Fos—dominant negative shown inhibit Jun-dependent transactivation prevent induction activity, only slightly basal activity promoter. Furthermore, A-Fos dominant phorbol ester Examination stable lines that overexpress revealed an Additionally, cisplatin-resistant line, A2780/CP70, which has compared parental A2780, have level. These results indicate can act may play role upregulation cancer cells.
