ABSTRACT
The H1 locus confers resistance to the potato cyst nematode Globodera rostochiensis pathotypes 1 and 4. It is positioned at distal end of chromosome V diploid Solanum tuberosum genotype SH83-92-488 (SH) on an introgression segment derived from S. ssp. andigena. Markers a high-resolution genetic map (Bakker et al. in Theor Appl Genet 109:146–152, 2004) were used screen BAC library construct physical covering 341-kb region resistant haplotype coming SH. For comparison, maps also generated two haplotypes susceptible RH89-039-16 (S. tuberosum/S. phureja), spanning syntenic regions 700 319 kb. Gene predictions genomic segments resulted identification large cluster consisting variable numbers CC-NB-LRR type R genes for each haplotype. Furthermore, interspersed with numerous transposable elements coding extensin-like protein amino acid transporter. Comparative analysis revealed major lack gene order conservation sequences three closely related haplotypes. Our data provide insight evolutionary mechanisms shaping will facilitate map-based cloning gene.
