ABSTRACT
The present study shows no adverse effects of the anti-diabetic drug metformin on bone mass and fracture healing in rodents but demonstrates that is not osteogenic vivo, as previously proposed. In view increased incidence fractures patients with type 2 diabetes mellitus (T2DM), we investigated metformin, a widely used T2DM therapy, vivo using two different rodent models modes administration. We first subjected 12-week-old female C57BL/6 mice to ovariectomy (OVX). Four weeks after OVX, received either saline or administered by gavage (100 mg/kg/daily). After 4 treatment, micro-architecture cellular activity were determined tibia micro-CT histomorphometry. another experiment, Wistar rats aged 3 months given only water for 8 via drinking (2 mg/ml). mid-diaphyseal osteotomy was performed left femur. Rats sacrificed architecture analysed right while callus volume femur X-ray analysis micro-CT, respectively. both models, our results show significant differences cortical trabecular metformin-treated compared saline. Metformin had effect resorption reduced formation rate bone. Mean scores assessed control showed between groups. Fracture mineral content similar Our indicate has rodents.
