ABSTRACT
The bacterium Escherichia coli is a well-studied recombinant host organism with plethora of applications in biotechnology. Highly valuable biopharmaceuticals, such as antibody fragments and growth factors, are currently being produced E. coli. However, the high metabolic burden during protein production can lead to cell death, consequent lysis, undesired product loss. Thus, fast precise analyzers monitor bioprocesses retrieve key process information, optimal time point harvest, needed. reliable monitoring tools still scarce date. In this study, we cultivated an strain producing single-chain fragment cytoplasm. bioreactor cultivations, purposely triggered lysis by pH ramps. We developed novel toolbox using UV chromatograms fingerprints chemometric techniques these events used flow cytometry (FCM) reference method quantify viability offline. Summarizing, were able show that comprising HPLC chromatogram fingerprinting data science allowed identification manner. convinced will not only facilitate bioprocess but also allow enhanced control future.
