ABSTRACT
Abstract In this study, both conventional one-dimensional liquid chromatography (1DLC) and comprehensive two-dimensional (2DLC) coupled to a high-resolution time-of-flight mass spectrometer (HR-TOF MS) were used for full-scale lipid characterization of extracts from zebrafish embryos. We investigated the influence on annotated lipids different separation mechanisms (HILIC, C18, PFP), their orders arranged in first second dimensions. As result, number species by LC-MS was between 212 448. contrast, individual C18×HILIC, HILIC×C18, HILIC×PFP 1784, 1059, 1123, respectively. Therefore, it evident that performance 2DLC, especially C18×HILIC method, considerably exceeded 1DLC. Interestingly, comparison HILIC×C18 approaches showed, under optimized conditions, similar orthogonality, but effective power much higher. A methods demonstrated had superior orthogonality with small increase its peak capacity, indicating combination maybe promising platform untargeted lipidomics complex samples. Finally, profiling respective, selected further studies.
