ABSTRACT
In the past decades, matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI) has been applied to a broad range of biological samples, e.g., forensics and preclinical samples. The use MALDI-MSI for analysis bone tissue limited due insulating properties material but more importantly absence proper sample preparation protocol undecalcified tissue. Undecalcified sections are preferred retain integrity as much possible or study tissue-bone bio interface in particular. Here, we optimized aimed at both targeted untargeted applications forensic applications, respectively. Different concentrations gelatin carboxymethyl cellulose (CMC) were tested embedding materials. composition 20% 7.5% CMC showed support best while sectioning. Bone be sectioned with tungsten carbide knife longitudinal fashion, need supported double-sided tapes maintain morphology developed sectioning method was shown applicable on rat mouse well human Targeted (methadone EDDP) (unknown lipids) detection demonstrated. DHB proved most suitable matrix methadone EDDP positive ion mode. limit (LOD) is estimated approximately 50 pg/spot successfully detect presence dosed femur clavicle. matrices unknown lipids hind legs mode CHCA based number tissue-specific peaks signal-to-noise ratios. method, animal bones, opens door future (pre)clinical investigations.
