ABSTRACT
Abstract Bacterial pathogens are fostered in and transmitted through wastewater. Hence, monitoring their impact on sanitation hygiene is imperative. As part of the process, culture-based methodologies primarily used, which centre use selective differential media. Media available today are, at best, difficult to formulate and, worst, prohibitively expensive. To address this lacuna, study proposes a medium for Klebsiella spp. blue agar (KBA) completely against selected gram-positive bacteria ( Bacillus spp., Staphylococcus aureus ) few gram-negative Acinetobacter baumanii , Serratia marcescens . On other hand, it supports growth chosen members pneumoniae species-complex with characteristic green colouration. Methylene blue, tryptophan, bile salt make up components KBA. Moreover, methylene 0.6% NaCl, glycerol render differential. KBA was more than HiCrome™ Selective Agar Base (KSA) replica plating experiments. promoted only 157 CFUs 209 KSA when stamped 253 grown LB. The colonies so isolated were predominantly identification colony polymerase chain reaction. nature distinguished aerogenes from species. contrary, lodged indistinguishable each Due its ease formulation, high selectivity, nature, cost-effective composition, viable option routine culture environmental clinical settings. Key points • Formulated novel media named Blue Facile formulation methodology Can be employed isolate complex sources such as wastewater
