ABSTRACT
To investigate the effects of photodynamic therapy (PDT) mediated by hematoporphyrin derivatives (HPD) on proliferation small cell lung cancer H446 cells and bronchial epithelial BEAS-2B cells. were cultured in vitro with different concentrations HPD(0, 5, 10, 12, 15, 20 μg/mL) for 4 h, then irradiated 630 nm laser energy densities (0, 25, 50, 75, 100 mW/cm2). Cell viability detected CCK8 assay. The apoptosis was observed Annexin V-FTTC/PI double staining Hoechst 33258. RT-PCR examination applied to detect transcriptional changes mRNA Bax、Bcl-2, Caspase-9. results showed that when HPD 15 μg/mL power density reached 50 mW/cm2, significantly decreased compared black control group. 33258 increase concentration, reduced, apoptotic increased. Flow cytometry assay revealed rates HPD-PDT group from those blank expression levels Bax Caspase-9 up-regulated, while Bcl-2 down-regulated significantly. can inhibit growth. mechanism may be related up-regulating down-regulating mRNA.
