ABSTRACT
Abstract Huang Qin decoction (HQD) is a traditional Chinese medicine formula for treating colitis, but the effects and molecular mechanism of action HQD in colitis-associated carcinogenesis (CAC) are still unclear. Therefore, we aimed to determine beneficial on CAC mice reveal underlying involved. AOM/DSS was used induce mice, tumorigenesis were examined (with mesalazine serving as positive control). Mesalazine or treatment alleviated body weight loss decreased disease activity index induced by AOM/DSS. also suppressed shortening colon tissue length, number tumors, infiltration inflammatory cells. The genes targeted predicted verified, followed knockout experiments. Elevated SLC6A4 inhibited serotonin production inflammation observed HQD-treated mice. NFκB NLRP3/caspase1/GSDMD pathways. therapeutic effect diminished SLC6A4-deficient Additionally, downregulation mitigated inhibitory HQD-containing serum MODE-K cell pyroptosis.
