Radio-synthesis and mass spectrometry analysis of 68Ga-DKFZ-PSMA-617 for non-invasive prostate cancer PET imaging

ABSTRACT
The focus of this paper in on quality control and decay property analysis of 68Ga-DKFZ-PSMA-617 for positron emission tomography (PET) imaging of prostate cancer. 68Ga-DKFZ-PSMA-617 was synthesized in high radiochemical yield and specific activity (77 GBq/µmol) under optimized conditions. The decayed tracer was characterized by MADLI-TOF to support its decay and coordination characterization. Contrast PET imaging studies were performed using normal nude mice after intravenous injection of 18.5 MBq of 68Ga-DKFZ-PSMA-617 and 18F-FDG respectively. These results warrant the further clinical application of 68Ga-DKFZ-PSMA-617 in prostate cancer PET imaging. 68Ga-DKFZ-PSMA-617 was synthesized in high radiochemical yield and specific activity (77 GBq/μmol) under optimized conditions. The corresponding decay tracer was firstly characterized by MADLI-TOF. Pre-clinical PET imaging studies were performed using normal nude mice after intravenous injection of 18.5 MBq of 68Ga-DKFZ-PSMA-617 which showed the typical enterohepatic metabolism. All these results warrant the further clinical application of 68Ga- DKFZ-PSMA-617.
