ABSTRACT
Abstract Background Currently, many micromammals are important targets for study. The endangered Galemys pyrenaicus is an outstanding example. Globally, their populations have suffered a substantial decline in last 20 years. In the surveyed area, capture of desman legally forbidden due to high conservation concerns. Reason by non-invasive sampling through faeces proposed its monitoring. Furthermore, confusion between from and Mediterranean water shrews must be considered. Thus, aim this study was focused on developing RT-PCR assays determine presence N. a. anomalus samples. Methods results conducted mountains System Central Extremadura (Spain). A total 186 samples were collected 2018 2021 experts where historically reported and/or our previous studies confirmed presence. using hydrolysis probes designed detect genetic material both specificity confirmed. reliability method further assessed PCR sequencing mitochondrial Cyb d-loop, resulting fully compatible with approach. Intraspecific phylogenetic relationship improve knowledge about mtDNA variability System. Conclusions We demonstrated that gives gold opportunity map species which minimizes disturbance reports population status individual Cost-effective combined field-collected allows us better investigate full range occurrence species.
