ABSTRACT
Potato is a member of the Solanaceae, plant family that includes several other economically important species, such as tomato, eggplant, petunia, tobacco and pepper. The Genome Sequencing Consortium (PGSC) aims to elucidate complete genome sequence potato, third most food crop in world. PGSC collaboration between 13 research groups from China, India, Poland, Russia, Netherlands, Ireland, Argentina, Brazil, Chile, Peru, USA, New Zealand UK. potato consists 12 chromosomes has (haploid) length approximately 840 million base pairs, making it medium-sized genome. sequencing project builds on diploid genomic bacterial artificial chromosome (BAC) clone library 78000 clones, which been fingerprinted aligned into ~7000 physical map contigs. In addition, BAC-ends have sequenced are publicly available. Approximately 30000 BACs anchored Ultra High Density genetic composed 10000 unique AFLPTM markers. From this integrated genetic-physical map, 50 150 seed currently identified for every chromosome. Fluorescent situ hybridization experiments selected BAC clones confirm these anchor points. provide starting point BAC-by-BAC strategy. This strategy being complemented by whole shotgun approaches using both 454 GS FLX Illumina GA2 instruments. Assembly annotation data will be performed available tailor-made tools. availability annotated help characterize germplasm collections based allelic variance assist breeders more fully exploit potential potato.
