ABSTRACT
Natural plant products have been widely used in controlling cancer with fewer or no side effects and the use of extracts as complementary to synthetic medicine is gaining increased popularity. Members Andrographis plants possess important medicinal properties. In present study, anti-cancerous properties nallamalayana (AN) were tested on A375 B16F10 skin melanoma cell lines. The leaf AN significantly reduced viability survival lines, achieved by MTT assay clonogenic assays, respectively. Further, TUNEL assays revealed that induces apoptosis. Western blot analysis expression Bcl-2, an anti-apoptotic protein induced proapoptotic molecules such Bcl-2 associated death promoter (BAD), X (BAX) cleaved caspase-3. Moreover, qRT-PCR western demonstrated G2/M phase proteins cdk1, cyclin B1 p53, cyclin-dependent kinase 1 inhibitor, p21. immunofluorescence NF-κB nuclear translocation, luciferase reporter gene activation. showed target genes. results concluded exhibited significant anti-proliferative, anti-inflammatory pro-apoptotic activities
