ABSTRACT
A new, more robust sprayer for desorption electrospray ionization (DESI) mass spectrometry imaging is presented. The main source of variability in DESI thought to be the uncontrolled various geometric parameters sprayer, primarily position solvent capillary, or specifically, its positioning within gas capillary nozzle. If off-center, becomes asymmetrical, making geometry difficult control and compromising reproducibility. stiffness, tip quality, are improved, reproducibility can improved by an order magnitude. quality potential high spatial resolution demonstrated on human colorectal tissue samples acquisition images at pixel sizes 100, 50, 20 μm, which corresponds a lateral 40–60 similar best values published literature. sensitivity also allows combination with fast scanning quadrupole time-of-flight spectrometer. This provides up 30 times faster acquisition, reducing overall time 10 mm × rat brain sample approximately 1 h. Although some spectral information lost increasing analysis speed, resulting data still used classify types basis previously constructed model. particularly interesting clinical applications, where fast, reliable diagnosis required.
